Cocaine, for example is active in the brain, and immune molecules don't cross the blood brain barrier. Cocaine molecules do, though. Nicotine has a more somatic response, but it is also a part of several rather important metabolic functions in the body, so basically, I have no idea how they plan to make reliable vaccines that actually work without making the patient sick....
